VIP (Vasoactive Intestinal Peptide) (Nasal Spray)

    Intranasal (nasal spray)4× daily

VIP (Vasoactive Intestinal Peptide) is a 28-amino acid neuropeptide with potent anti-inflammatory action. It is used intranasally in the Shoemaker protocol for CIRS.

Reviewed by the AllPeptides.eu Editorial Team·Last reviewed: 2026-06-16

    Mechanism of Action

VIP acts via VPAC1/VPAC2 receptors, reducing inflammatory cytokines (TNF-α, IL-6), regulating T-regulatory cells, and improving pulmonary function. The intranasal route allows for local action on the nasal mucosa and potential central access via the olfactory nerve.

    Overview

    • Nasal administration — no injection
    • Dosing: 50 mcg per spray, 4×/day (Shoemaker protocol)
    • Use only after completing steps 1–11 of the Shoemaker protocol
    • No reconstitution required — ready-to-use formulation

    Intranasal Delivery — Research Data

    • VIP is administered intranasally, primarily within the framework of the Shoemaker protocol for CIRS (Chronic Inflammatory Response Syndrome).
    • The nasal bioavailability of VIP is not fully documented, but clinical experience supports the use of this route.
    • Extremely sensitive peptide — requires strict refrigeration and careful handling.

    Limited evidence

Very few published data on intranasal delivery.

Shoemaker Protocol: use exclusively after completing CIRS steps (1–11). Start low due to risk of hypotension. Self-compounding instructions are not provided.

    Formulation Notes

    No self-preparation instructions provided

    • Available as a ready-to-use nasal spray from specialized compounding pharmacies.
    • Self-compounding a nasal spray is not recommended due to the peptide's sensitivity.
    • Typical concentration: 50 mcg per spray.

    Storage After Opening

    • Refrigerate (2–8 °C) after opening.
    • Use within 14 days.
    • Ideally: freeze in aliquots for longer shelf life.

    Dosing Protocol

    • Use only after completing the Shoemaker protocol (steps 1–11).
    • Start with 50 mcg (1 spray/nostril), 4×/day.
    • Increase to 100 mcg (2 sprays/nostril) after 2 weeks if well tolerated.
    • Cycle: 30 days or as per medical guidance.

    Storage Instructions

    • Refrigerate (2–8 °C) — mandatory.
    • Protect from light and heat.
    • Use within 14 days after opening.

    Important Notes

    • Use ONLY after completion of the Shoemaker protocol (CIRS).
    • May cause hypotension — start with a low dose.
    • Extremely sensitive peptide — avoid room temperature.
    • Self-compounding instructions are not provided.
