Kisspeptin (5mg)
Kisspeptin (also known as metastin) is a naturally occurring neuroendocrine peptide that plays a central role in human reproduction by stimulating GnRH secretion. It binds to the GPR54 receptor in the hypothalamus, triggering pulsatile GnRH release and subsequent LH and FSH secretion. This protocol outlines a once-daily subcutaneous approach.

How It Works
Kisspeptin is an upstream trigger of the reproductive hormone cascade. After subcutaneous injection, Kisspeptin rapidly binds to GPR54 receptors on hypothalamic GnRH neurons, inducing an immediate release of GnRH. This, in turn, stimulates the anterior pituitary gland to secrete LH and FSH, which act on the gonads to enhance sex steroid production and support gametogenesis. Kisspeptin's action is GnRH-dependent—if GnRH release is blocked, Kisspeptin cannot induce LH/FSH secretion. This provides a more physiological pattern of hormone release.
Quick Start
- Reconstitution: Add 1.5 mL of bacteriostatic water → ~3.33 mg/mL concentration.
- Typical daily range: 100–200 mcg once daily (gradual titration).
- Storage: Lyophilized: freezer −20 °C; after reconstitution, refrigerator 2–8 °C; avoid freeze-thaw cycles.
Typical / Gradual Approach (1.5 mL = ~3.33 mg/mL)
Route: Subcutaneously | Frequency: Once daily
| Week | Dose | Units (per injection) |
|---|---|---|
| Weeks 1–2 | 100 mcg | 3 Units (0.03 mL) |
| Weeks 3–8 (or 3–12) | 200 mcg | 6 Units (0.06 mL) |
Studies report administration once daily subcutaneously. Conservative titration to assess individual response.

Reconstitution Steps
- Draw up 1.5 mL of bacteriostatic water.
- Inject slowly down the side of the vial; avoid foaming.
- Gently swirl/rotate until dissolved (do not shake).
- Label and refrigerate at 2–8 °C, protecting from light.
Protocol Overview
- Goal: Support of physiological reproductive hormone signaling via upstream GnRH stimulation.
- Schedule: Daily subcutaneous injections for 8–12 weeks.
- Storage: Lyophilized frozen; reconstituted refrigerated; avoid freeze-thaw cycles.
Dosing Protocol
- Initiation: 100 mcg daily for 1–2 weeks to assess sensitivity.
- Titration: Increase to 200 mcg daily if well-tolerated.
- Frequency: Once daily, subcutaneously.
- Cycle Duration: 8–12 weeks; avoid prolonged continuous use due to tachyphylaxis.
- Timing: Consistent time; rotate sites.
- Break between cycles: After a cycle (8–12 weeks), a 4–6 week pause is recommended. Kisspeptin can cause tachyphylaxis—the pause restores GPR54 sensitivity.
Washout & Cycle Restart
When can I restart?
After a cycle (8–12 weeks), a 4–6 week pause is recommended. Kisspeptin can cause tachyphylaxis—the pause restores GPR54 sensitivity.
Minimum wait period: 4 weeks
Storage Instructions
- Lyophilized: Store at −20 °C in dry, dark conditions.
- Reconstituted: Refrigerate at 2–8 °C; stable for up to ~4 weeks; avoid freeze-thaw cycles.
- Let vials reach room temperature before opening.
Important Notes
- Discard bacteriostatic water 28 days after opening.
Potential Benefits & Side Effects
Potential Benefits:
- Physiological sex hormone stimulation: Increases endogenous testosterone/estrogen via LH/FSH without HPG suppression.
- Fertility restoration: Shows promise in hypothalamic amenorrhea.
- IVF ovulation trigger: May reduce risk of ovarian hyperstimulation syndrome (OHSS).
- Safety profile: Generally well-tolerated; occasional mild injection site reactions.
Potential Side Effects:
- Injection site reactions (redness, pain).
- Hot flashes/flushing (due to GnRH activation).
- Headache (transient).
- Nausea (rare).
- Abdominal discomfort (rare).
⚠️ Serious Warnings & Long-Term Risks
The following warnings are based on published literature, FDA labels, and post-marketing surveillance. They do not constitute medical advice. Consult a healthcare professional.
HPG axis dysregulation with chronic use
Kisspeptin regulates the GnRH pulse generator. Chronic exogenous administration may theoretically dysregulate the reproductive axis (tachyphylaxis).
