DSIP (5mg)
Delta Sleep-Inducing Peptide (DSIP) is a naturally occurring nonapeptide (9 amino acids) originally isolated from rabbit brain tissue and is studied for its effects on sleep architecture and stress regulation. Research suggests DSIP may promote delta wave (slow-wave) sleep, regulate cortisol and ACTH levels, and exhibit stress-protective properties. This protocol outlines a once-daily subcutaneous approach.

How It Works
DSIP (Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu) was first characterized in 1977 as a sleep-regulating factor. Mechanistic studies suggest it may influence sleep via modulation of GABAergic transmission and interaction with opioid receptor systems. Research indicates it can increase the duration of delta wave (slow-wave) sleep without significantly altering REM sleep architecture. Beyond sleep, DSIP has shown stress-protective and adaptogenic properties through modulation of the hypothalamic-pituitary-adrenal axis.
Quick Start
- Reconstitution: Add 3.0 mL bacteriostatic water → ~1.67 mg/mL concentration.
- Typical Daily Range: 100–300 mcg once daily (gradual titration); advanced up to 500 mcg.
- Storage: Lyophilized: freeze −20 °C; after reconstitution, refrigerate 2–8 °C; avoid freeze-thaw.
Typical / Gradual Approach (3 mL = ~1.67 mg/mL)
Route: Subcutaneously | Frequency: Once daily
| Week | Dose | Units (per injection) |
|---|---|---|
| Week 1 | 100 mcg | 6 Units (0.06 mL) |
| Week 2 | 150 mcg | 9 Units (0.09 mL) |
| Week 3 | 200 mcg | 12 Units (0.12 mL) |
| Weeks 4–8 | 250–300 mcg | 15–18 Units (0.15–0.18 mL) |
Studies report administration once daily subcutaneously, typically in the evening before bed.

Reconstitution Steps
- Aspirate 3.0 mL of bacteriostatic water.
- Inject slowly down the wall; avoid frothing.
- Gently swirl/rotate until dissolved (do not shake).
- Label and refrigerate at 2–8 °C, protecting from light.
Protocol Overview
- Goal: Support healthy sleep architecture and stress regulation.
- Schedule: Daily subcutaneous injections for 4–8 weeks (extendable to 12 weeks).
- Storage: Lyophilized frozen; reconstituted refrigerated; avoid freeze-thaw.
Dosing Protocol
- Initiation: 100 mcg daily; increase by ~50 mcg every 1–2 weeks.
- Target: 250–300 mcg daily by Weeks 4–8.
- Frequency: Once daily (subcutaneously), typically before bed.
- Cycle Duration: 4–8 weeks; optional extension to 12 weeks with periodic breaks.
- Break between cycles: After the cycle (4–6 weeks), a 2–4 week washout period is recommended. DSIP modulates sleep — the pause prevents reliance on an exogenous sleep peptide.
Washout & Cycle Restart
When can I restart?
After the cycle (4–6 weeks), a 2–4 week washout period is recommended. DSIP modulates sleep — the pause prevents reliance on an exogenous sleep peptide.
Minimum wait period: 2 weeks
Storage Instructions
- Lyophilized: Store at −20 °C in dry, dark conditions.
- Reconstituted: Refrigerate 2–8 °C; use within ~4 weeks; avoid freeze-thaw.
- Allow vials to reach room temperature before opening.
Important Notes
- Some research suggests DSIP effects may persist for multiple nights; adjust frequency if needed.
Potential Benefits & Side Effects
Potential Benefits:
- May promote deeper, restorative slow-wave sleep and improve subjective sleep quality.
- Research suggests potential stress-protective and anxiolytic effects.
- Some studies indicate DSIP does not induce pharmacological tolerance with continuous use.
- Generally well-tolerated in human studies.
Potential Side Effects:
- Occasional mild injection site reactions.
- Limited long-term safety data; periodic breaks are recommended.
⚠️ Serious Warnings & Long-Term Risks
The following warnings are based on published literature, FDA labels, and post-marketing surveillance. They do not constitute medical advice. Consult a healthcare professional.
Hormonal dysregulation
DSIP modifies cortisol, LH, GH. Long-term use may dysregulate the HPA axis. Very few human studies.
