DSIP (10mg)
DSIP dosing protocols can help promote deep, restorative delta-wave sleep and support a healthy stress response. Delta Sleep-Inducing Peptide (DSIP) is a naturally occurring nonapeptide (9 amino acids) originally isolated from rabbit brains. Research suggests it may improve sleep quality, normalize sleep architecture, and support mood stabilization without next-day drowsiness. Research sources report subcutaneous administration of 100 mcg to 300 mcg, once daily, at bedtime, increased gradually.

How It Works
DSIP acts as a neuromodulator of the sleep-wake cycle and stress response. Administration consistently increases delta-wave (slow-wave) sleep on EEG recordings, suggesting it promotes the onset and maintenance of deep, non-REM sleep. DSIP readily crosses the blood-brain barrier for central effects and may enhance inhibitory neuronal activity. Additionally, it mitigates ACTH and corticosterone release in response to stress, indicating an effect on the hypothalamic-pituitary-adrenal (HPA) axis. Unlike conventional sedatives, it tends to normalize sleep architecture without next-day drowsiness.
Quick Start
- Reconstitution: Add 3.0 mL bacteriostatic water → 3.33 mg/mL concentration.
- Range reported in research sources: 100 mcg to 300 mcg once daily, at bedtime, increased gradually.
- Easy measurement: At 3.33 mg/mL, 300 mcg = 9 units (0.09 mL) on a U-100 syringe.
- Storage: Lyophilized: freeze at −20 °C; after reconstitution, refrigerate at 2–8 °C; avoid freeze-thaw cycles.
Ranges reported in research sources (3.0 mL = 3.33 mg/mL)
Route: Subcutaneously | Frequency: Once daily, at bedtime
| Week | Dose | Units (per injection) |
|---|---|---|
| Week 1 | 100 mcg | 3 Units (0.03 mL) |
| Week 2 | 150 mcg | 4.5 Units (0.04 mL) |
| Week 3 | 200 mcg | 6 Units (0.06 mL) |
| Weeks 4–8 | 250–300 mcg | 7.5–9 Units (0.07–0.09 mL) |
Research sources report subcutaneous administration once daily, at bedtime, increased gradually from 100 mcg to 300 mcg. The values are calculation examples from research sources, not validated human dosing.

Reconstitution Steps
- Withdraw 3.0 mL of bacteriostatic water.
- Inject slowly down the side wall; avoid frothing.
- Gently swirl/rotate until dissolved (do not shake).
- Label and refrigerate at 2–8 °C, protecting from light.
Protocol Overview
- Goal: Support improved sleep quality, enhanced delta-wave sleep, and stress regulation.
- Storage: Lyophilized frozen; reconstituted refrigerated; avoid freeze-thaw cycles.
- Schedule reported in research sources: 100 mcg to 300 mcg subcutaneously, once daily, at bedtime, increased gradually over 8 weeks.
- Vials for the full schedule: about 2 vials of 10 mg (≈13 mg in total).
Dosing Protocol
- Week 1: 100 mcg once daily, at bedtime.
- Week 2: 150 mcg once daily, at bedtime.
- Week 3: 200 mcg once daily, at bedtime.
- Weeks 4–8: 250–300 mcg once daily, at bedtime.
- Cycle Duration: 8–12 weeks; consider a post-cycle break.
- There is no validated human dosing — research context only.
- Break between cycles: After the 8-week schedule, sources describe a break of 2–4 weeks.
Washout & Cycle Restart
When can I restart?
After the 8-week schedule, sources describe a break of 2–4 weeks.
Minimum wait period: 2 weeks
Storage Instructions
- Lyophilized: Store at −20 °C in dry, dark conditions.
- Reconstituted: Refrigerate 2–8 °C; prepare aliquots if needed; avoid freeze-thaw cycles.
- Allow vials to reach room temperature before opening.
Important Notes
- Avoid other sedatives while using DSIP to clearly assess sleep outcomes.
Potential Benefits & Side Effects
Potential Benefits:
- Supports improved sleep quality and increased duration of slow-wave (delta) sleep.
- May help normalize disturbed sleep patterns in chronic insomnia.
- Shows potential for stress reduction and mood support via HPA axis regulation.
- Notably safe profile: mild, transient side effects (occasional headache or nausea).
Potential Side Effects:
- Injection site reactions (redness, itching).
- Drowsiness or extended sleep (expected).
- Headache (rare).
- Mild dizziness post-administration.
- Limited long-term safety data.
⚠️ Serious Warnings & Long-Term Risks
The following warnings are based on published literature, FDA labels, and post-marketing surveillance. They do not constitute medical advice. Consult a healthcare professional.
Hormonal dysregulation
DSIP modifies cortisol, LH, GH. Long-term use may dysregulate the HPA axis. Very few human studies.
