CJC-1295 DAC (5mg)
CJC-1295 DAC is a long-acting synthetic GHRH analog. The DAC (Drug Affinity Complex) modification reversibly binds to albumin, extending its half-life to ~6–8 days. By stimulating pituitary GHRH receptors, it increases GH secretion and circulating IGF-1 in a dose-dependent manner. This protocol outlines a twice-weekly subcutaneous approach.

How It Works
CJC-1295 DAC mimics endogenous GHRH, binding to GHRH receptors on pituitary somatotroph cells to release GH. The DAC (albumin-binding moiety) modification keeps the GHRH analog in circulation for days instead of minutes, extending its half-life to ~6–8 days. The increase in GH stimulates IGF-1 production (primarily in the liver); IGF-1 mediates many developmental and metabolic effects via JAK/STAT. Pulsatile GH secretion is still preserved during continuous CJC-1295 stimulation.
Quick Start
- Reconstitution: Add 2.0 mL bacteriostatic water → 2.5 mg/mL (2500 mcg/mL) concentration.
- Typical Dose Range: 300–1,000 mcg per injection, twice weekly (gradual titration).
- Storage: Lyophilized: freezer -20 °C; after reconstitution, refrigerator 2–8 °C; avoid freeze-thaw.
Typical / Gradual Approach (2 mL = 2500 mcg/mL)
Route: Subcutaneous | Frequency: Twice-weekly
| Week | Dose | Units (per injection) |
|---|---|---|
| Weeks 1–2 | 300 mcg (0.3 mg) | 12 Units (0.12 mL) |
| Weeks 3–4 | 500 mcg (0.5 mg) | 20 Units (0.20 mL) |
| Weeks 5–6 | 750 mcg (0.75 mg) | 30 Units (0.30 mL) |
| Weeks 7–12 | 1,000 mcg (1 mg) | 40 Units (0.40 mL) |
Studies report administration twice-weekly subcutaneously (e.g., Monday/Thursday). The extended 6–8 day half-life due to DAC supports less frequent dosing. Weekly dosing is also studied.

Reconstitution Steps
- Withdraw 2.0 mL bacteriostatic water.
- Inject slowly down the side of the vial; avoid foaming.
- Gently swirl/rotate until dissolved.
- Label and refrigerate at 2–8 °C, protected from light.
Protocol Overview
- Objective: Support sustained elevation of GH and IGF-1 for research on GH axis effects.
- Schedule: Twice-weekly subcutaneous injections for 8–12 weeks (extendable to 16).
- Storage: Lyophilized frozen; reconstituted refrigerated.
Dosing Protocol
- Initiation: 300 mcg per injection twice/wk; increase by ~250 mcg every 2 weeks.
- Target: 750–1,000 mcg per injection by weeks 5–12.
- Frequency: Twice-weekly (subcutaneous), with 3–4 days between injections.
- Cycle Duration: 8–12 weeks; optional extension to 16.
- Timing: Consistent time of day (e.g., evening); rotate sites.
- Break between cycles: After the cycle, a washout period of 4–8 weeks is recommended. CJC-1295 DAC has a prolonged half-life (6–8 days)—the break allows for recovery of the GHRH axis.
Washout & Cycle Restart
When can I restart?
After the cycle, a washout period of 4–8 weeks is recommended. CJC-1295 DAC has a prolonged half-life (6–8 days)—the break allows for recovery of the GHRH axis.
Minimum wait period: 4 weeks
Storage Instructions
- Lyophilized: Store at -20 °C in dry, dark conditions.
- Reconstituted: Refrigerate 2–8 °C; stable for 2–4 weeks; avoid freeze-thaw cycles.
- Allow to reach room temperature before opening.
Important Notes
- Log each dose, site, and timing.
- The DAC modification provides sustained release; maintain a consistent twice/wk schedule.
Potential Benefits & Side Effects
Potential Benefits:
- Supports sustained, dose-dependent increases in GH and IGF-1.
- May promote increased lean mass, reduced fat mass, and improved body composition.
- Enhanced protein synthesis and recovery potential via anabolic signaling.
- Generally well-tolerated; mild injection site reactions.
- Some report transient flushing, headache, or water retention during initial titration.
Potential Side Effects:
- Flushing / sensation of warmth (common, transient).
- Headache (transient).
- Fluid retention (mild edema, especially initially).
- Injection site reactions (redness, tenderness).
- Numbness/tingling (rare, GH-related).
⚠️ Serious Warnings & Long-Term Risks
The following warnings are based on published literature, FDA labels, and post-marketing surveillance. They do not constitute medical advice. Consult a healthcare professional.
Theoretical risk (prolonged GH elevation)
The DAC variant provides continuous GH elevation for 7–10 days. Theoretical concern for IGF-1-related oncogenesis.
