    How It Works

5-Amino-1MQ inhibits the NNMT enzyme in adipocytes → increased intracellular NAD+ → activation of SIRT1/SIRT3 sirtuins → increased mitochondrial β-oxidation of fatty acids + browning of white adipose tissue. Concurrently, it reduces SAM-dependent methylation associated with lipogenesis.

    Quick Start

    • Form: 50mg capsule — no reconstitution required.
    • Typical research dose: 50–150 mg daily, divided into 1–3 doses.
    • Administration time: Morning (or split morning/midday), with or without food.
    • Research stage: Preclinical — limited human data.

Route: Oral (per os) | Frequency: 1–3 times daily

    WeekDoseAmount
    Weeks 1–2
    50 mg (50,000 mcg)
    1 x 50mg capsule (morning)
    Weeks 3–4
    100 mg (100,000 mcg)
    2 x 50mg capsules (morning + midday)
    Weeks 5–8
    150 mg (150,000 mcg)
    3 x 50mg capsules (morning + midday + afternoon)

Dosing is extrapolated from preclinical data. Split the dose into 1–3 administrations per day. The small molecule profile supports oral bioavailability.

    Protocol Overview

    • 5-Amino-1MQ is a small molecule inhibitor of NNMT — an enzyme that converts nicotinamide to N-methylnicotinamide, reducing NAD+ levels.
    • NNMT inhibition → increased NAD+ → activation of SIRT1/SIRT3 → increased mitochondrial function in adipocytes.
    • In mice on a high-fat diet, administration of 5-Amino-1MQ reduced body weight by ~7% and induced browning of white adipocytes (browning effect) without changing food intake (Neelakantan et al., 2018).

    Dosing Protocol

    • Start with 50mg daily (1 capsule in the morning) for the first 2 weeks.
    • Increase to 100mg (morning + midday) for weeks 3–4.
    • If well-tolerated, increase to 150mg daily (3 capsules).
    • An 8-week cycle is typically followed by a 4-week break.
    • Break between cycles: A 4-week break between cycles is recommended.

    Washout & Cycle Restart

    When can I restart?

A 4-week break between cycles is recommended.

    Minimum wait period: 4 weeks

    Storage Instructions

    • Store at room temperature (15–25 °C).
    • Away from moisture and light.
    • Keep sealed between uses.

    Important Notes

    • ⚠️ Human clinical data is very limited.
    • NNMT inhibition may affect nicotinamide metabolism — monitor B3/NAD+ levels.
    • Not an approved medication — research compound only.
    • Avoid during pregnancy/breastfeeding.
