Thymosin Alpha-1 (5 mg) & MOTS-C (5 mg) (Stack)
Thymosin Alpha-1 (Tα1, a thymic immunomodulator approved in 30+ countries) and MOTS-C (a mitochondrial metabolic peptide, AMPK) are combined for immuno-metabolic support: Tα1 enhances T-cells/NK cells, MOTS-C improves metabolism/energy.

How It Works
**Tα1** activates dendritic cells, increases CD4+/CD8+ T-cells, regulates NK cells, and enhances phagocytic capacity. **MOTS-C** activates AMPK by inhibiting the folate cycle, enhancing glucose uptake, fatty acid oxidation, and mitochondrial respiration. The combination: Tα1 → immunological enhancement + MOTS-C → metabolic optimization, targeting the immune-metabolism connection.
Quick Start
- Tα1: Add 2.5 mL BAC water → 2.0 mg/mL.
- MOTS-C: Add 1.0 mL BAC water → 5 mg/mL (1 vial = 1 dose).
- Tα1: 0.9–1.6 mg 2×/week subcutaneously.
- MOTS-C: 5 mg 2–3×/week subcutaneously.
- Storage: Tα1: 2–8 °C, 28 days. MOTS-C: 2–8 °C, 28 days.
Tα1 (2×/wk) & MOTS-C (2–3×/wk) Dosing
Route: Subcutaneous | Frequency: Tα1: 2×/week | MOTS-C: 2–3×/week
| Week | Dose | Units (per injection) |
|---|---|---|
| — Thymosin Alpha-1 — | ||
| Weeks 1–4 | 0.9 mg (2×/wk) | 45 units (0.45 mL) |
| Weeks 5–8 | 1.6 mg (2×/wk) | 80 units (0.80 mL) |
| — MOTS-C — | ||
| Weeks 1–2 | 5 mg | 100 units (1.00 mL) × 2/wk |
| Weeks 3–8 | 5 mg | 100 units (1.00 mL) × 3/wk |
Tα1 is administered 2 times/week (e.g., Monday/Thursday). MOTS-C 2–3×/week. They can be injected on the same days at different injection sites.

⚠️ Units change depending on BAC water — always verify before administration. Based on
- Tα1: Withdraw 2.5 mL BAC water → 2.0 mg/mL.
- MOTS-C: Withdraw 1.0 mL BAC water → 5 mg/mL.
- Label the peptide & date; refrigerate at 2–8 °C.
Protocol Overview
- Objective: Immuno-metabolic optimization — enhancing immune function + metabolism.
- Tα1: T-cells, NK cells, dendritic cells. Approved as Zadaxin®.
- MOTS-C: AMPK activation, insulin sensitivity, endurance.
Dosing Protocol
- Tα1: 0.9 mg × 2/wk (wks 1–4) → 1.6 mg × 2/wk (wks 5–8).
- MOTS-C: 5 mg × 2/wk (Wks 1–2) → 5 mg × 3/wk (Wks 3–8).
- Break between cycles: After the cycle, a washout period of 4–6 weeks is recommended. Tα1 regulates the immune system and MOTS-C regulates mitochondria — the pause prevents overstimulation/tolerance.
Washout & Cycle Restart
When can I restart?
After the cycle, a washout period of 4–6 weeks is recommended. Tα1 regulates the immune system and MOTS-C regulates mitochondria — the pause prevents overstimulation/tolerance.
Minimum wait period: 4 weeks
Storage Instructions
- Tα1: Lyophilized −20 °C; reconstituted 2–8 °C within 28 days.
- MOTS-C: Lyophilized −20 °C; reconstituted 2–8 °C within 28 days.
Important Notes
- Tα1: use with caution in autoimmune diseases.
- MOTS-C: ongoing clinical trials — human data is limited.
Potential Benefits & Side Effects
Potential Benefits:
- Tα1: Enhancement of T/NK cells, antiviral activity, immunostimulation.
- MOTS-C: Insulin sensitivity, fat reduction, increased endurance.
- Combination: Holistic immuno-metabolic support.
Potential Side Effects:
- Tα1: mild injection site redness, very rarely fever.
- MOTS-C: none reported in preclinical studies.
⚠️ Serious Warnings & Long-Term Risks
The following warnings are based on published literature, FDA labels, and post-marketing surveillance. They do not constitute medical advice. Consult a healthcare professional.
Immunostimulation — risk in autoimmune conditions
Tα1 enhances cellular immunity (CD4+, CD8+, NK). Theoretical risk of autoimmune flare (MS, RA, Lupus). Contraindicated in active autoimmune disease.
📄 Expert Opin Biol Ther 2009AMPK regulation — effect on glucose metabolism
MOTS-c activates AMPK and modifies glucose metabolism. Combined with antidiabetic drugs, may theoretically cause hypoglycemia. Very few human studies.
