How It Works
**FOXO4-DRI** is a D-retro-inverso peptide that competes with the natural FOXO4 protein. In senescent cells, FOXO4 binds to p53, preventing their apoptosis. FOXO4-DRI disrupts this bond, freeing p53, which activates mitochondrial apoptosis. **Epithalon** (Ala-Glu-Asp-Gly) activates the gene expression of hTERT (telomerase), elongating telomeres and increasing the Hayflick limit (number of cell divisions).

Protocol Overview
- FOXO4-DRI (D-Retro-Inverso) is a designer peptide developed at Erasmus MC (Netherlands, 2017). It disrupts the FOXO4-p53 axis, forcing senescent cells into apoptosis.
- Epithalon (AEDG peptide) was developed by Professor Vladimir Khavinson. It activates telomerase (hTERT), elongating telomeres.
- The combination: removal of senescent cells + telomere renewal = a holistic anti-aging approach.
Dosing Protocol
- FOXO4-DRI: 5 mg subcutaneously every 14 days × 3 doses (total of 6 weeks).
- Epithalon: 5 mg subcutaneously, 1x/day × 10–20 days (in the evening before sleep).
- Cycle: 2 times/year (every 6 months).
- They can be started simultaneously, or Epithalon can be started after the last FOXO4-DRI pulse.
- Break between cycles: 6-month pause between cycles. Senolysis is pulse-based and does not require continuous administration.

Important Notes
- FOXO4-DRI has not been tested in human clinical trials — only preclinical data (mice) is available.
- Do not administer to patients undergoing chemotherapy — senolysis may interact.
- Epithalon has 15+ years of clinical data (Russia).
- Bloodwork (telomeres via TeloYears or Life Length) is recommended before and after the cycle.

Telomerase & oncogenesis — conflicting data
Epithalon activates telomerase (hTERT). While it appeared anti-cancer in animal models, telomerase activation is considered a hallmark of cancer. No long-term human studies exist.
