How It Works
The combination targets three fundamental mechanisms of aging: **Epithalon** activates telomerase (hTERT), elongating telomeres and reversing cellular senescence (Hayflick limit). **MOTS-C** activates the AMPK → SIRT1 axis, optimizes mitochondrial metabolism, and regulates glucose. **SS-31** binds to cardiolipin, stabilizes complexes I–IV of the respiratory chain, reduces ROS by 40–60%, and prevents mitochondrial dysfunction. Together, they act on DNA (telomeres), metabolism (AMPK), and energy production (ETC) — the three pillars of biological aging.

- Epithalon 10mg: Add 2ml of bacteriostatic water → 5mg/ml. Swirl gently.
- MOTS-C 10mg: Add 2ml of bacteriostatic water → 5mg/ml. Swirl gently.
- SS-31 10mg: Add 2ml of bacteriostatic water → 5mg/ml. Swirl gently.
- Do not use NaCl — only bacteriostatic water (benzyl alcohol 0.9%).
- Allow the powder to dissolve naturally — do not shake violently.
Protocol Overview
- This stack targets the three central mechanisms of aging simultaneously:
- Epithalon (Epitalon) — A tetrapeptide (Ala-Glu-Asp-Gly) that activates telomerase, elongating telomeres and slowing cellular senescence. Discovered by Prof. Vladimir Khavinson.
- MOTS-C — A mitochondrial peptide (16 amino acids) that regulates glucose metabolism, enhances AMPK, and improves insulin sensitivity. Encoded by mitochondrial DNA.
- SS-31 (Elamipretide) — A tetrapeptide (D-Arg-Dmt-Lys-Phe) that binds to cardiolipin on the inner mitochondrial membrane, stabilizing the electron transport chain.
- The synergy: Epithalon protects at the chromosomal level, MOTS-C optimizes mitochondrial function, and SS-31 prevents mitochondrial damage.
Dosing Protocol
- Wk. 1 — Introduction: Epithalon 5mg SC in the evening, MOTS-C 5mg SC in the morning (3x/wk), SS-31 2.5mg SC in the morning.
- Wk. 2 — Full Dose: Increase SS-31 to 5mg/day. Epithalon and MOTS-C remain unchanged.
- Wk. 3–6 — Maintenance: Continue with full doses. MOTS-C is ideally taken before exercise.
- Epithalon is administered in the evening — melatonin enhances its action on the pineal gland.
- MOTS-C can be administered on exercise days for synergy via AMPK.
- Monitor energy levels, sleep quality, and exercise endurance.
- Break between cycles: After the cycle (4–6 weeks), a washout period of 8–12 weeks is recommended. Epithalon activates telomerase — the pause prevents chronic overstimulation. MOTS-C and SS-31 act at the mitochondrial level — the pause allows for assessment of results and natural homeostasis.
Washout & Cycle Restart
When can I restart?
After the cycle (4–6 weeks), a washout period of 8–12 weeks is recommended. Epithalon activates telomerase — the pause prevents chronic overstimulation. MOTS-C and SS-31 act at the mitochondrial level — the pause allows for assessment of results and natural homeostasis.

Storage Instructions
- All peptides (powder): -20°C for long-term storage, 2–8°C for short-term.
- After reconstitution: 2–8°C (refrigerator), away from light.
- Epithalon: Use within 28 days after reconstitution.
- MOTS-C: Use within 21 days after reconstitution.
- SS-31: Use within 21 days after reconstitution.
- Do not leave out of the refrigerator for more than 30 minutes.

- Investigational stack — no clinical studies exist for this triple combination.
- Epithalon has been researched for over 30 years (Khavinson Institute) with positive safety data.
- MOTS-C may lower glucose — caution is advised for diabetics on medication.

Telomerase & oncogenesis — conflicting data
Epithalon activates telomerase (hTERT). While it appeared anti-cancer in animal models, telomerase activation is considered a hallmark of cancer. No long-term human studies exist.

MOTS-c activates AMPK and modifies glucose metabolism. Combined with antidiabetic drugs, may theoretically cause hypoglycemia. Very few human studies.
