Triptorelin (GnRH) (2mg)
Triptorelin is a synthetic decapeptide analog of gonadotropin-releasing hormone (GnRH). In micro-doses (50–100 mcg), it is used in research to restart the hypothalamic-pituitary-gonadal (HPG) axis. CRITICAL WARNING: Clinical doses in mg (3.75–11.25 mg) cause chemical castration — PCT protocols use mcg only.

How It Works
Triptorelin (D-Trp6-GnRH) is a synthetic decapeptide GnRH analog with a D-tryptophan substitution at position 6, which dramatically increases stability and potency (100× more potent than natural GnRH). In **micro-doses** (50–100 mcg), it acts as an acute GnRH agonist, causing a transient release of LH and FSH from the anterior pituitary. This 'wakes up' the HPG axis that was suppressed by exogenous substances. LH stimulates the Leydig cells of the testes to produce testosterone, while FSH supports spermatogenesis. **At high doses** (mg), continuous occupation of GnRH receptors causes downregulation via receptor internalization, leading to complete LH/FSH suppression — chemical castration. This is why dosing accuracy is of vital importance.
Quick Start
- Reconstitution: Add 2.0 mL bacteriostatic water → 1 mg/mL (1000 mcg/mL).
- PCT Dose: Single injection of 100 mcg (NOT mg) — 'one-shot PCT'.
- ⚠️ CRITICAL: 100 mcg ≠ 100 mg. Wrong unit = chemical castration. TRIPLE-check.
PCT Micro-Dose (2 mL = 1 mg/mL)
Route: Intramuscular or Subcutaneous | Frequency: One-time (single shot) or weekly micro-dose
| Week | Dose | Units (per injection) |
|---|---|---|
| One-Shot PCT | 100 mcg (0.1 mg) | 10 Units (0.10 mL) |
| Low-Dose PCT (alt.) | 50 mcg (0.05 mg) | 5 Units (0.05 mL) |
One-Shot PCT: A single injection of 100 mcg after complete clearance of exogenous substances. Low-Dose: 50 mcg once, often combined with a SERM (Tamoxifen/Clomid) for 14–28 days. ⚠️ DO NOT use mg doses.

Reconstitution Steps
- Withdraw 2.0 mL of bacteriostatic water.
- Inject slowly against the wall · do not spray directly onto the powder.
- Gently swirl until fully dissolved — do not shake.
- Note the concentration (1 mg/mL = 1000 mcg/mL) and date.
- Refrigerate at 2–8 °C · use within 28 days.
Protocol Overview
- Goal: Restart the HPG axis after an anabolic or exogenous testosterone cycle.
- Schedule: A single 100 mcg injection ('one-shot PCT') or a single 50 mcg injection + SERM.
- CRITICAL: Doses in mg (3.75+ mg) are used in prostate cancer therapy and cause chemical castration.
- Timing: Inject ONLY after exogenous compounds have fully cleared.
Dosing Protocol
- One-Shot PCT: 100 mcg intramuscularly or subcutaneously — single injection.
- Low-Dose PCT: 50 mcg single injection + Tamoxifen 20mg/day or Clomid 25–50mg/day × 14–28 days.
- Timing: After full clearance of exogenous substances — depends on half-life.
- ⚠️ DO NOT repeat within 4 weeks — triptorelin has a potent effect on the HPG axis.
- Break between cycles: After injection, wait 4–6 weeks before evaluating your hormone panel. Do not use again without laboratory assessment.
Washout & Cycle Restart
When can I restart?
After injection, wait 4–6 weeks before evaluating your hormone panel. Do not use again without laboratory assessment.
Minimum wait period: 4 weeks
Storage Instructions
- Lyophilized: −20 °C in dark, dry conditions.
- Reconstituted: Refrigerate 2–8 °C · use within 28 days.
- Avoid freeze-thaw cycles.
Important Notes
- ⚠️ CRITICAL DOSING WARNING: 100 mcg (micrograms), NOT 100 mg. Doses in mg cause chemical castration.
- Triptorelin is ~100× more potent than natural GnRH.
- Do not use if exogenous substances have not fully cleared.
- Single use — does not require cycles or repetitions in most cases.
- Check hormone panel (LH, FSH, testosterone) 4–6 weeks after.
Potential Benefits & Side Effects
Potential Benefits:
- Rapid HPG axis restart in a 'one-shot' protocol.
- More potent and targeted than Gonadorelin due to the D-Trp6 modification.
- Simplicity: a single injection instead of weekly PCT regimens.
Potential Side Effects:
- Overdose = chemical castration: The most significant concern. Doses in mg completely suppress testosterone.
- Transient 'flare': a temporary increase in LH/testosterone before potential stabilization.
- Possible headache, hot flashes, or mood changes after injection.
- Rare injection site reactions.
⚠️ Serious Warnings & Long-Term Risks
The following warnings are based on published literature, FDA labels, and post-marketing surveillance. They do not constitute medical advice. Consult a healthcare professional.
Complete gonadotropin suppression (chemical castration)
Triptorelin is a GnRH agonist that after initial flare-up completely suppresses LH/FSH, testosterone, and estradiol. Causes reversible chemical castration. Requires medical supervision.
📄 FDA Label — Trelstar®Cardiovascular risk due to hypogonadism
Androgen deprivation therapy (ADT) with GnRH agonists is associated with increased risk of MI, stroke, and QT prolongation.
