    How It Works

Semaglutide is a GLP-1 agonist with 94% homology to human GLP-1. It reduces appetite, slows gastric emptying, and improves glycemic control. Its half-life is ~7 days due to albumin binding.

    Quick Start

    • Reconstitution: Add 2.0 mL bacteriostatic water → 5.0 mg/mL concentration.
    • Weekly range: 0.25–2.4 mg once weekly.
    • Storage: Lyophilized: −20 °C; reconstituted: 2–8 °C, within 28 days.

Studies report administration once weekly on the same day. The higher concentration (5 mg/mL) reduces injection volumes by half compared to the 5 mg vial. One 10 mg vial is sufficient for ~4 weeks of maintenance at 2.4 mg/week.

    Reconstitution Steps

    1. Draw 2.0 mL of bacteriostatic water.
    2. Inject slowly down the side of the vial; avoid foaming.
    3. Swirl gently until dissolved.
    4. Note the date; refrigerate at 2–8 °C.
    5. Use within 28 days.

    Protocol Overview

    • Objective: Chronic weight management via GLP-1 activation.
    • Schedule: Weekly injections for 16–20+ weeks.

    Dosing Protocol

    • Wk 1–4: 0.25 mg/wk.
    • Wk 5–8: 0.5 mg/wk.
    • Wk 9–12: 1.0 mg/wk.
    • Wk 13–16: 1.7 mg/wk.
    • Wk 17+: 2.4 mg/wk (maintenance).
    • Break between cycles: After completing a cycle, a 4–8 week washout period is recommended before repeating. Semaglutide has a long half-life (~7 days)—the body needs time to fully clear the peptide and restore natural GLP-1 regulation.

    Washout & Cycle Restart

    When can I restart?

After completing a cycle, a 4–8 week washout period is recommended before repeating. Semaglutide has a long half-life (~7 days)—the body needs time to fully clear the peptide and restore natural GLP-1 regulation.

    Minimum wait period: 4 weeks

    Storage Instructions

    • Lyophilized: −20 °C.
    • Reconstituted: 2–8 °C; use within 28 days; do not freeze.

    Important Notes

    • If a dose is missed, take it within 5 days.
    • Gradual titration reduces GI side effects.
