GHRP-2 (10mg)
GHRP-2 dosing protocols utilize this potent synthetic hexapeptide (pralmorelin) that stimulates growth hormone (GH) release by acting as a ghrelin receptor agonist in the pituitary and hypothalamus. Researchers have explored GHRP-2 as a diagnostic GH stimulant and for supporting healthy GH secretion patterns, lean body composition, recovery, and sleep quality. This protocol outlines a once-daily subcutaneous approach.

How It Works
GHRP-2 (pralmorelin) binds to the ghrelin/GHS receptor (GHS-R1a) on pituitary somatotropes and in the hypothalamus. By activating these receptors, it triggers a strong pulse of GH release, independent of GHRH. Research shows it can stimulate GH even when GHRH action is impaired. Unlike exogenous GH, it maintains natural pulsatile secretion patterns and works synergistically with endogenous GHRH.
Quick Start
- Reconstitution: Add 3.0 mL bacteriostatic water → ~3.33 mg/mL concentration.
- Typical Daily Range: 100–300 mcg once daily (gradual titration).
- Timing: Evening administration before sleep; on an empty stomach preferred.
- Storage: Lyophilized: freeze −20 °C; after reconstitution, refrigerate 2–8 °C; use within 28 days.
Standard / Gradual Approach (3 mL = ~3.33 mg/mL)
Route: Subcutaneous | Frequency: Once Daily (Evening)
| Week | Dose | Units (per injection) |
|---|---|---|
| Weeks 1–2 | 100 mcg | 3 Units (0.03 mL) |
| Weeks 3–4 | 150 mcg | 4.5 Units (0.04 mL) |
| Weeks 5–8 | 200 mcg | 6 Units (0.06 mL) |
| Weeks 9–12 | 250–300 mcg | 7.5–9 Units (0.07–0.09 mL) |
Studies report administration once daily subcutaneously before bed on an empty stomach (at least 2–3 hours after eating).

Reconstitution Steps
- Withdraw 3.0 mL of bacteriostatic water.
- Inject slowly down the side of the vial; avoid foaming.
- Gently swirl/rotate until dissolved (do not shake).
- Note date and refrigerate at 2–8 °C; use within 28 days.
Protocol Overview
- Goal: Support pulsatile GH release for body composition, recovery, and sleep quality.
- Schedule: Daily subcutaneous injections for 8–16 weeks.
- Storage: Lyophilized frozen; reconstituted refrigerated; avoid freeze-thaw cycles.
Dosing Protocol
- Initiation: 100 mcg daily; increase by ~50 mcg every 1–2 weeks.
- Target: 200–300 mcg daily by Wk. 5–12.
- Frequency: Once daily (subcutaneous) before bed.
- Cycle Duration: 8–12 weeks; optional extension to 16 weeks.
- Timing: Before bed on an empty stomach; rotate sites.
- Break between cycles: A 4–6 week washout period is recommended post-cycle. GHRP-2 can cause GHSR tolerance — a washout restores sensitivity.
Washout & Cycle Restart
When can I restart?
A 4–6 week washout period is recommended post-cycle. GHRP-2 can cause GHSR tolerance — a washout restores sensitivity.
Minimum wait period: 4 weeks
Storage Instructions
- Lyophilized: Store at −20 °C; stable for years.
- Reconstituted: Refrigerate 2–8 °C; use within 28 days; avoid freeze-thaw cycles.
- Allow vials to reach room temperature before opening.
Important Notes
- Reconstituted vials and opened bacteriostatic water must be discarded after 28 days.
Potential Benefits & Side Effects
Potential Benefits:
- Supports pulsatile GH release and may increase IGF-1 levels with consistent use.
- May support improvements in body composition, recovery, and sleep quality.
- Transient increases in appetite (ghrelin receptor activation), cortisol, and prolactin.
- Generally well-tolerated; occasional mild injection site reactions.
Potential Side Effects:
- Occasional mild injection site reactions (redness/itching).
- Receptor desensitization (tachyphylaxis) can occur with continuous daily use; cyclic protocols may help.
⚠️ Serious Warnings & Long-Term Risks
The following warnings are based on published literature, FDA labels, and post-marketing surveillance. They do not constitute medical advice. Consult a healthcare professional.
Increased cortisol & prolactin (less than GHRP-6)
Less ghrelin-hunger than GHRP-6, but still increases cortisol. Long-term effects on HPA axis unclear.
