Adamax (10mg)
Adamax is an adamantane-modified analogue of Semax (N-acetyl Semax amide), a heptapeptide derivative of ACTH(4–10) studied for its nootropic and neuroprotective properties. The adamantane group enhances blood-brain barrier passage and peptide stability, yielding greater potency than standard Semax. This protocol outlines a once-daily subcutaneous approach.

How It Works
Adamax is structurally derived from Semax, an an ACTH(4–10) analogue with nootropic properties. Semax increases the expression of BDNF (Brain-Derived Neurotrophic Factor) and enhances TrkB receptor sensitivity in the hippocampus, promoting neuroplasticity, synaptic growth, and neuronal survival. The peptide modulates neurotransmitter systems (dopamine, serotonin) and activates intracellular pathways (cAMP/CREB). The adamantane group of Adamax confers greater lipophilicity and enzymatic stability, allowing for more efficient BBB penetration. Preclinical data suggest a 2–3 fold greater efficacy on cognitive outcomes compared to standard Semax.
Quick Start
- Reconstitution: Add 3.0 mL bacteriostatic water → ~3.33 mg/mL concentration.
- Typical daily range: 500–1000 µg once daily (gradual titration).
- Storage: Lyophilized: −20 °C; after reconstitution, refrigerate 2–8 °C; use within 1–2 weeks; avoid freeze-thaw cycles.
Standard / Titration Approach (3 mL = ~3.33 mg/mL)
Route: Subcutaneously | Frequency: Once daily
| Week | Dose | Units (per injection) |
|---|---|---|
| Weeks 1–2 | 300 µg (0.3 mg) | 9 Units (0.09 mL) |
| Weeks 3–4 | 500 µg (0.5 mg) | 15 Units (0.15 mL) |
| Weeks 5–6 | 750 µg (0.75 mg) | 22.5 Units (0.22 mL) |
| Weeks 7–8 | 1000 µg (1.0 mg) | 30 Units (0.30 mL) |
Studies report administration once daily subcutaneously.

Reconstitution Steps
- Draw up 3.0 mL of bacteriostatic water.
- Inject slowly down the side of the vial; avoid creating foam.
- Gently swirl/rotate until fully dissolved (do not shake vigorously).
- Note the date and refrigerate at 2–8 °C, protected from light.
Protocol Overview
- Objective: Support cognitive enhancement, neuroprotection, and neuroplasticity via BDNF regulation.
- Schedule: Daily subcutaneous injections for 8–12 weeks (extendable to 16), followed by an equal off-cycle period.
- Storage: Lyophilized frozen; reconstituted refrigerated; avoid freeze-thaw cycles.
Dosing Protocol
- Initiation: 300 µg daily for 2 weeks.
- Titration: 500 µg (Wk 3–4), then 750 µg (Wk 5–6).
- Maintenance: 1000 µg daily (Wk 7+).
- Frequency: Once daily, preferably in the morning.
- Cycle: 8–12 weeks; equal time off-cycle to prevent tolerance.
- Break between cycles: After the cycle (8–12 weeks), a washout period of 4–8 weeks is recommended. Adamax increases BDNF — an equal off-cycle period prevents tolerance.
Washout & Cycle Restart
When can I restart?
After the cycle (8–12 weeks), a washout period of 4–8 weeks is recommended. Adamax increases BDNF — an equal off-cycle period prevents tolerance.
Minimum wait period: 4 weeks
Storage Instructions
- Lyophilized: Store at −20 °C in dry, dark conditions.
- Reconstituted: Refrigerate at 2–8 °C; use within 1–2 weeks.
- Avoid Freeze-Thaw: Do not re-freeze reconstituted solution.
- Allow vial to reach room temperature before opening.
