| Mechanism context | Ipamorelin binds to the growth hormone secretagogue receptor (GHSR-1a) and stimulates the pituitary to release endogenous growth hormone in a pulsatile manner. Unlike older GH-releasing peptides, it is highly selective and does not significantly stimulate ACTH, cortisol or prolactin at the studied doses. After subcutaneous injection, GH levels peak at ~40 minutes and return to baseline within 2-3 hours. This short-acting pulsatile action makes it suitable for daily administration. Animal studies showed that even chronic daily exposure did not desensitize the GH release mechanisms. Ipamorelin has also shown prokinetic effects on the gastrointestinal system. | GHRP-6 acts as a synthetic ghrelin mimetic binding to the growth hormone secretagogue receptor (GHS-R1a) in the pituitary gland and hypothalamus. This activation triggers acute, pulsatile release of growth hormone from somatotroph cells while simultaneously reducing the inhibitory somatostatin brake on GH secretion. Unlike continuous GH administration, GHRP-6 preserves physiological feedback controls. Beyond endocrine effects, it exhibits cytoprotective properties through interactions with the CD36 receptor on immune and muscle cells. |
