Mechanism contextCJC-1295 DAC is a modified 30-amino-acid GHRH analogue conjugated to a Drug Affinity Complex (DAC) that binds covalently to serum albumin after injection. This albumin binding dramatically extends the half-life from minutes to approximately 6-8 days, allowing weekly dosing. It binds to GHRH receptors on pituitary somatotroph cells, enhancing GH synthesis and pulsatile release. Importantly, it increases basal GH levels while preserving the natural GH pulse rhythm. Clinical trials show sustained GH increases of 2-10 fold and IGF-1 of 1.5-3 fold, with elevated IGF-1 persisting up to 28 days after multiple doses.Mod GRF 1-29 stimulates the GHRH receptor in the pituitary, activating pulsatile growth hormone secretion. Unlike CJC-1295 DAC, its short half-life preserves the physiological pulsatile pattern.
