    What is PNC-27 and how does it work?

    PNC-27 is a chimeric 32-amino-acid peptide joining the post-activation segment of p53 (residues 12–26) with a cell penetration signal. In tumor models, it binds membrane HDM2/MDM2, assembles transmembrane pores, and induces rapid necrosis, while largely sparing normal cells.

    Selective toxicity against cancer cells

    The key characteristic of PNC-27 is its selective action: it targets cells expressing HDM2 on their membrane — something observed mainly in certain cancer cell lines. Normal cells, which do not express membrane HDM2, are not significantly affected in the same preclinical models.

    Pore formation and necrosis mechanism

    Microscopy studies show PNC-27 forms toroidal pores in cancer cell membranes after binding membrane HDM2. These pores disrupt membrane integrity, leading to rapid cell death through necrosis rather than apoptosis.

    Regulatory framework and warnings

    PNC-27 is not approved by any regulatory agency for therapeutic use. It has triggered warnings regarding unapproved use in patients. Research remains exclusively at the preclinical stage in cell lines and animal models.
