    What is Mazdutide?

    Mazdutide (IBI-362 / LY3305677) is a weekly dual GLP-1 and glucagon receptor agonist, designed based on oxyntomodulin. In phase 2 and 3 studies, Mazdutide produced clinically significant weight loss along with improvements in multiple metabolic markers. China granted the first marketing approvals in 2025.

    Mechanism of action

    Acting simultaneously on GLP-1 and glucagon receptors, Mazdutide reduces appetite (GLP-1) while increasing energy expenditure (glucagon). This dual mechanism theoretically offers advantages over GLP-1 mono-agonists like semaglutide.

    Clinical results

    In randomized clinical trials in Chinese adults with overweight/obesity, Mazdutide achieved mean weight loss exceeding 10% at 24 weeks. Additionally, it improved glycemic control, blood lipids, and liver function markers.

    Comparison with Semaglutide and Tirzepatide

    While semaglutide (Ozempic/Wegovy) is a GLP-1 mono-agonist and tirzepatide (Mounjaro) is a dual GLP-1/GIP, Mazdutide targets GLP-1 and glucagon. The glucagon component theoretically increases energy metabolism, but head-to-head trials are needed for definitive comparisons.
